DEK Protein
Source: GST-tagged recombinant human kidney produced in E. coli.
Purity: > 90% by SDS-PAGE, apparent Mr ~ 70-kDa.
Supplied: 1 μg in 50 μl 50 mM Tris-HCl pH 7.0 buffer containing 14 mM β-mercaptoethanol, 1 mM benzamidine, 0.1 mM phenylmethanesulfonyl fluoride, 1 mM EDTA and 10% glycerol. Maintain preparations in aliquots at -70° C. Avoid repeated thawing.

Background: DEK is a nuclear phosphoprotein of apparent Mr ~ 43 kDa that is fused to the nucleoporin CAN as a result of a recurrent (6;9) chromosomal translocation in a subset of acute myeloid leukemias and myelodysplastic syndrome. DEK also acts as an autoimmune antigen in juvenile rheumatoid arthritis and other rheumatic diseases. DEK associates with the transcriptional co-repressor Human Daxx, the SRm160 splicing coactivator and the HIV-2 enhancer. It changes the topology of DNA by introducing constrained positive supercoils into protein-free and chromatin-associated DNA. DEK exhibits some structural similarity to I2PP2A/SET, which also occurs as a fusion protein with CAN in acute undifferentiated leukemia. However, DEK does directly affect the activity of PP2A.
